Nhân sâm là một trong những thành phần quan trọng nhất trong các phương thuốc thảo dược Đông Á. Hiện nay, nhân sâm được bổ sung vào các sản phẩm bổ sung khác nhau như chiết xuất dạng lỏng, nước uống  tăng  lực,  trà, viên nang, viên nén,…Theo dược điển Mỹ USP 40  th́ì  trong các sản phẩm nhân sâm các ginsenoside Rb1, Rg1, Rf, Re là các thành phần hoạt tính chính được tìm thấy. Nhằm góp phần vào hoạt động kiểm tra chất lượng của sản phẩm mà trong công bố có bổ sung sâm, công tác kiểm nghiệm các ginsenoside là rất cần thiết.

Nghiên cứu nhằm mục tiêu định lượng ginsenoside Rb1, Rg1, Rf, Re trong thực phẩm bổ sung bằng kỹ thuật sắc ký lỏng ghép khối phổ hai lần (LC-MS/MS).

Các ginsenoside được chiết bằng dung dịch n-butanol bão hòa nước sau khi loại béo bằng diethyl ether, loại carbohydrate bằng nước, định lượng bằng HPLC-MS/MS; triển khai quy trình định lượng các ginsenoside trong thực phẩm bổ sung. Quy trình xác định ginsenoside Rb1, Rg1, Rf, Re bằng máy sắc ký lỏng ghép khối phổ hai lần (LC-MS/MS) trong nền mẫu cao sâm, nước uống bổ sung và viên nang uống bổ sung.

Kết quả nghiên cứu cho thấy, hoàn thành quy trình định lượng các ginsenoside Rb1, Rg1, Rf, Re trong thực phẩm bổ sung bằng phương pháp LC-MS/MS với giới hạn phát hiện là 0,5 µg/mL, giới hạn định lượng là 2,0 µg/mL, độ chụm RSD% <20%, hiệu suất thu hồi trong khoảng 80 – 110%. Ứng dụng quy trình phân tích để định lượng các ginsenoside trong 48 mẫu thực phẩm bổ sung trên thị trường. Sắc ký lỏng ghép khối phổ hai lần là phương pháp có độ nhạy cao, thích hợp cho định lượng các ginsenoside trong thực phẩm bổ sung có nền mẫu phức tạp.
